Cytotoxicity testing evaluates the toxic effects of compounds on living cells. At GISB Lab LLC, USA our cytotoxicity testing services support pharmaceutical screening, medical device evaluation, and raw material quality control. Whether you need qualitative or quantitative cytotoxicity tests, our GLP-compliant laboratory delivers accurate, reliable results.
Cytotoxicity testing is essential before extensive toxicological studies. Our lab performs both qualitative assays and quantitative analysis using validated methods per USP and ISO standards.

Standards & Compliance
- USP <87> Biological Reactivity Tests, In Vitro
- ISO 10993-5 Cytotoxicity Testing
- GLP Compliant Testing
Cytotoxicity Testing Methods
Direct Contact Cytotoxicity Test
The direct contact test places the test article directly on L-929 mouse fibroblast cells. After 24-hour incubation, cells are examined microscopically and graded 0-4 based on reactivity. This test method is ideal for solid materials of normal density.
Agar Diffusion Cytotoxicity Test (Agarose Overlay)
The agar diffusion test uses an agarose layer between the test sample and cells. This testing method protects cells from physical damage, making it preferred for high-density or low-density materials. The response is scored on a 0-4 scale after 24+ hours.
MEM Elution Cytotoxicity Test
The MEM elution cytotoxicity test evaluates extractables and leachables from the test article. The extract is applied to L-929 cells for 48 hours. This testing approach detects toxic substances that may leach from materials during use.
MTT Assay – Quantitative Cytotoxicity Testing
The MTT assay is a quantitative test measuring cell viability via mitochondrial activity. Living cells convert MTT dye to purple formazan; dead cells cannot. Our MTT testing evaluates extracts at multiple concentrations (undiluted, 1:2, 1:4, 1:10) and quantifies viable cell percentage using spectrophotometry.
The MTT assay provides objective, reproducible cytotoxicity data superior to qualitative grading methods. Choose MTT testing for sensitive detection of cytotoxic effects.
